Affilin molecules represent a new class of so-called scaffold proteins, artificial proteins designed to selectively bind antigens. Affilin proteins are structurally derived from human ubiquitin (and historically from gamma-B crystallin). The concept of scaffold proteins is to use stable and versatile protein structures that can confer de novo binding properties and specificity by introducing mutations in surface-exposed amino acid residues. The construction of affilin phage display libraries can be used to identify novel protein ligands. Complex variants and combinations are generated by randomized genetic methods, resulting in large cDNA libraries. Candidates that bind to the desired target can be selected by display methods, especially phage display, which is very robust and flexible. Affilin proteins are constructed by modifying the surface-exposed amino acids of these proteins and isolated by display techniques such as phage display and screening. They are also similar to antibodies in their affinity and specificity for antigen, but differ in structure, making them an antibody mimetic.
