Rabbits have been used to produce polyclonal antibodies because they are larger than mice and can collect more antibodies from their serum. However, some antigens cannot induce an immune response in mammals, especially those that are highly conserved. Birds and mammals diverged from each other more than 300 million years ago. This evolutionary time interval reduces the similarity between even highly conserved proteins, thereby increasing antigenicity. Therefore, proteins that cannot trigger an immune response in mice or rabbits usually cause a strong response in chickens. All vertebrates have similar immune mechanisms, so chicken antibodies can play a role in failed mammalian applications. In addition, chickens produce much more antibodies than mice by secreting antibodies against eggs. Phage display allows rapid selection of monoclonal antibodies without being limited by traditional hybridoma technology. However, the application of phage display technology in the production of chicken monoclonal antibodies is limited. To avoid this defect, we developed a special technology combined with phage display to apply in chicken monoclonal antibody production, which is more robust and much less biased in clone selection.
