DARPins Ready-to-panning Phage Display Library Construction

DARPins (designed ankyrin repeat proteins) are genetically engineered antibody-mimicking proteins that typically exhibit highly specific and high-affinity binding to target proteins. They are derived from native ankyrin repeat proteins, the most common class of binding proteins in nature, and are responsible for a variety of functions such as cell signaling, regulation, and cellular structural integrity. DARPins consist of at least three repeating motifs or modules, of which the most N-terminal and the most C-terminal modules are called "caps" because they shield the protein's hydrophobic core. DARPins libraries are designed by sequence alignment of thousands of native ankyrin repeat motifs (~33 amino acids each) combined with structure-based design and recombinant DNA methods. These proteins are composed of repeating building blocks that form a stable protein domain with a large potential target interaction surface. DARPins correspond to the average size of native ankyrin repeat protein domains. Proteins with fewer than three repeats (i.e., capping repeats and one internal repeat) do not form sufficiently stable tertiary structures.
