Direct Isolation of PhageThe direct isolation method of bacteriophages is to extract bacteriophages from microorganisms and particulate matter in solid or liquid environmental samples. Then, through plaque detection, the extracted sample is used to infect the host bacteria. This method provides a snapshot of all phages present in the environmental sample that may infect the host. The combination of direct isolation and plaque assay is a rapid method for detecting phages in solid and liquid environmental samples. In fact, using direct isolation, you may find many different phages with different plaque morphologies. However, because not every phage in the sample can successfully infect the host bacteria, you may find as few as one plaque on the plate or no plaque at all.   • This method extracts bacteriophages from microorganisms and particulate matter in solid or liquid environmental samples. And this method provides a snapshot of all phages present in the environmental sample that may infect the host.

  • Plaque assay
  • The plaque assay allows you to visually confirm the presence of phage particles in the sample.

Direct Isolation Procedure

  • Prepare workbench for aseptic work and assemble your supplies
  • Collect environmental samples according to your needs.
  • Extract bacteriophages from solid environmental samples (such as soil or compost). If your collection is a liquid sample, we will proceed directly to next step
  • Prepare phage filtrate using the aseptic technique.
