Enriched Isolation of Phage

The bacteriophage is first extracted from a solid environmental sample into a bacterial growth medium. Filter to remove bacteria, including those that are potentially dangerous. The filtered sample is inoculated with host bacteria, and the culture is cultivated to promote growth and replication. As the bacteria replicate, the phages present in the sample undergo multiple rounds of infection and lysis. The result is that the concentration of bacterial host-specific phage in the culture is exponentially higher than the initial environmental sample. Enrichment and separation are achieved by mixing soil samples with a bacterial growth medium to extract bacteriophages and then filtering out unwanted soil microorganisms. The bacterial host cells are then added to the filtrate. Any phage particle that recognizes a specific host can be grown and amplified through multiple rounds of lytic growth. A single phage particle can multiply into millions of particles. It is important to note that a particular type of phage may outperform other phages that are present, so the phage collection at the end may not necessarily represent the phage present in starting sample.
