The plaque count is considered the gold standard for phage enumeration. A single plaque is the result of a bacteriophage initially lysing a host bacterium, followed by subsequent bacteriophage killing neighboring cells. Conversely, the larger area of clearance may be the result of other antibacterial compounds or more phages mixed in the sample. The dual agar overlay assay (DLA) allows localized phage-host contact in a closed environment (petri dish) containing two layers of agar. The bottom layer is prepared with a bacterial growth-supporting medium containing 1-1.5% agar. The top layer contains the same medium, but with a lower concentration of agar, which is usually referred to as soft-agar and it is mixed with host bacteria and poured on the bottom layer to form a so-called lawn. If the phage can infect the tested bacteria, there will be obvious spots or plaques on the lawn, which contain lysed bacteria and phage particles. If the phage does not form clearly visible plaques, the agar may be replaced by agarose. The concentration (titer) of infectious phage in the phage stock solution or sample can be determined by spotting serially diluted samples on the bacterial lawn.
