Enumeration and Detection of Whole Phage Particles

Compared with traditional optical microscopes, transmission electron microscopes (TEM) use electron beams to produce 1000 times the resolution. The increased resolution (down to 0.2 nm) is sufficient to visualize the virus. This technique can be used to quantify phage particles, although the sample needs to be highly concentrated (~106 particles/mL) to produce reliable results. Phage quantification using TEM is very accurate in determining the morphology and total number, but it is considered time-consuming, expensive, and impractical, unable to run many samples, and cannot be used for complex samples. In addition, sample preparation is cumbersome, and the technique requires skilled operators on complex instruments. Another technique for calculating the entire particle is flow cytometry. Here, the virus particles are labeled with fluorescent dyes and guided through the capillary. The small diameter of the capillary forces the particles to flow in a line, allowing detection of the light scattering caused by each particle. This method is fast and rigorous, so it is widely used.
