Fab Phage Display System ConstructionPhage display technology is to clone the polypeptide or protein-coding gene or target gene fragment into the appropriate position of the phage coat protein structural gene so that the exogenous polypeptide or protein and coat protein are fused and expressed, and the fusion protein is displayed in the recombination of progeny phage. The phage vector pComb3 has been widely used in the construction of the natural phage antibody library and the expression of Fab antibody. The construction of the phage vector pComb3 is a fusion protein formed by linking the Fd chain of the antibody to the C-terminal region of the phage coat protein III (pIII). The Fd/pIII fusion protein and light chain protein genes are under the control of the sequence of the Lac promoter/controller, respectively, and are translated into proteins and then introduced into the bacterial cell membrane space. The heavy chain Fd is embedded in the membrane by fusion with pIII, while the light chain is secreted alone into the intermembrane space. The heavy and light chains assemble on the membrane to form active Fab antibodies.
