Hyperphage Display System Construction

Hyperphages carry the pIII gene deletion. They are produced by an E. coli packaging cell line that produces a functional pIII for packaging phage genomes with pIII deletions. The resulting hyperphages carry functional pIII on their surface but lack the pIII gene in their genome. These hyperphages can then be used to infect bacteria with phagemid libraries. Each resulting display phage carries multiple copies of the antibody or peptide on its surface, significantly increasing the panning efficiency. A helper phage technology ("Hyperphage System") has been developed by Rondot et al. Hyperphage system allows for improved antibody presentation in phage display by increasing the number of antibodies displayed per phage particle (up to 5 vs. 0.01), so this system offers great advantages in functional gene analysis and proteomics. Phage panning can be performed with small amounts of antigen and with greater efficiency. For example, the use of universal libraries for antibody isolation can be improved by panning hyperphage libraries on Western blots following two-dimensional gel electrophoresis.

Hyperphage System Application

Large numbers of open reading frames (ORFs) can be analyzed by panning against synthetic membrane-bound peptide epitopes. In cancer research, hyperphage libraries can be a tool to discover new tumor markers by translating them to the cell surface.
