Phage-Derived Depolymerases ProductionIn addition to specific antibiotic resistance, bacterial biofilm formation can lead to another level of complications in attempts to eradicate pathogenic or harmful bacteria, including difficulty in penetration of drugs into bacterial cells through biofilm structures, compromised host immune responses, and Accumulation of various biologically active substances. Depolymerases are phage-derived enzymes that facilitate the early stages of phage infection by degrading extracellular material that encapsulates bacteria and may also help reach phage receptors. Depolymerase activity is particularly interesting in removing biofilms as it alters the EPS matrix and reduces bacterial virulence. They are able to degrade capsular polysaccharides, exopolysaccharides and O-polysaccharide chains from lipopolysaccharides and peptidoglycans. All of these substances may constitute the envelope of some free-living bacteria, but they are mostly important components of the biofilm matrix. Depolymerases can bind to virions, form part of the phage particle, or be in a soluble form. Since phage-encoded depolymerases are capable of degrading polysaccharides in bacterial capsules and biofilm matrices, phages encoding these enzymes may be more readily accessible to bacterial hosts, leading to infection.

Phage-Derived Depolymerases Applications

  • Phage depolymerases as an alternative to antibiotics in the treatment of multidrug-resistant bacteria
    Since the discovery of the activity of phage depolymerases, they have become an alternative to antibiotics, especially in the treatment of multidrug-resistant bacteria. Unlike antibiotics, depolymerases can specifically target the host and allow the natural bacterial flora to remain intact. Not only can they be engineered to increase their degradative activity, but they can also be used as a tool to detect bacteria.
  • Combination therapy with phage-derived depolymerases and different therapeutic agents to disrupt bacterial biofilms
    Using phage-encoded depolymerases as the only therapeutic approach to disrupt bacterial biofilms is not sufficient. Phage depolymerases sometimes only reduce biofilm biomass by about 20%. Therefore, combination therapy is recommended. This method combines two or more agents simultaneously or sequentially. Combination therapy consists of some phage-derived depolymerases and different alternative therapeutics, including antibiotics, phages, chemical or natural compounds, and detergents.
