Proteases are an important class of enzymes whose activity is critical for many physiological and pathological processes. These enzymes are involved in a variety of biological functions, including tissue remodeling and morphogenesis, infection, coagulation, neoplasia, and cancer metastasis. Protease activity can serve as a valuable diagnostic biomarker and therapeutic target. Given their importance and diversity, there is an unmet need for unbiased techniques for the independent analysis of protease activity and as components of complex biological mixtures. Only a small subset of the physiological substrates of proteases have been characterized in some detail. Currently, unbiased protease analysis methods are based on mass spectrometry and tend to be cumbersome and expensive. Targeted activity-based assay techniques can detect active proteases, but are typically limited to low levels of assay multiplexing and are limited to enzymes with well-characterized substrates. At Creative Biolabs, our team used phage display of random peptide libraries to characterize cleavage motifs to overcome these problems.
