The classic method is to incubate the solid-phase or liquid-phase antigen and antibody libraries. After several rounds of "adsorption-elution-amplification", specific antibodies can be enriched. Creative Biolabs specializes in the production of phage display libraries for antibodies obtained from immune sources. Our most common request is a VHH single domain antibody library from immunized alpacas or llamas. We can assist in the procurement of antigens and immunization strategies and library creation and phage display library screening. The deliverable can be simple sequence information of the selected antibody or a small amount of soluble purified antibody. Our phage display vector and display system are unique and have many advantages. We have also made custom scFv libraries from mice and humans, and can make various other sources (rabbit, sheep, cow, etc), as long as antibody sequence information is available. We can build the binding domain into the Fc backbone or other configurations, and perform various optimization procedures when necessary. Phage Display Steps

  • Panning
    • Panning is an iterative process of enriching bacteriophages in a population. Compared with other populations, this population has high affinity binding to the target of interest.
  • Colony selection
    • Then phages that selected from the previous step are cloned and picked to isolate each unique protein-binding agent.
  • Ag-antibody interaction
    • During panning, phage displaying proteins with higher binding affinity are selected relative to phage displaying proteins with lower affinity
  • Function screening
  • After characterizing the antibody-antigen interaction, cell-based detection methods are often used to screen candidate molecules for functional activity.
