Phage Display Peptide Library Screening

Peptides are two-dimensional linear amino acid chains, usually very short in length (less than 50 AA). They are either designed through a reasonable calculation method, or through phage display screening to obtain peptides that highly specifically bind to the target, and may modulate the target. Compared with antibodies (~150 kDa), peptides are relatively small (~3–5 kDa), so they are easy to synthesize and modify, have higher cell membrane permeability, and have lower immunogenicity. In cancer treatment, these peptides can be used as targeting ligands to help specifically deliver cytotoxic drugs to the tumor vasculature, tumor microenvironment, or cancer cells. On the other hand, peptides can also be delivered intracellularly to target cancer to specifically upregulate transcription factors, oncogenes, or enzymes. Phage display is a powerful technique for screening and isolating target-specific peptides. This method uses phage to display foreign peptides or antibodies on its surface by inserting genes encoding corresponding polypeptides into the phage genome.
