Unlike natural and immune libraries, semi-synthetic libraries consist of a mixture of natural and chemically synthesized sequences. Gene synthesis methods are commonly used to provide random complementarity determining region (CDR) combinations. The advantage of semi-synthetic antibody libraries is that the framework sequences to be used are predetermined. This will help the development and application of specific antibodies. The semi-synthetic library is carefully designed by assuming the number of scaffolds, the positions to be diversified, the types of amino acids to be included in the design, and the ratio of amino acids to be diversified in each position. These assumptions are not always correct. In order to avoid making assumptions about the structure of the CDR, a combination of synthetic scaffolds and natural CDRs was used.
