Phage DNA Vaccines

The phage DNA vaccine consists of a eukaryotic expression cassette that encodes a specific antigen controlled by a target-specific promoter contained in the phage. The expression cassette must have the necessary regulatory sequences to allow correct gene expression and protein folding of the antigen. In this strategy, phage particles act as passive vectors, allowing the DNA-encoded antigen to be transferred to eukaryotic cells, where the DNA is expressed. Phage DNA vaccines are considered to be more advantageous than standard DNA vaccines because the former is more stable in storage, transportation, and administration (mainly via oral routes) because the capsid of the phage particles protects the DNA from degradation. Although ʎ phage is the most common phage vector used for DNA vaccination, filamentous phages have also been tested. An important feature to consider for filamentous phage-DNA vaccines is that the phagemid vector can effectively contain multiple gene copies, which may allow the use of a single delivery vector to immunize various epitopes or antigens. Compared with the naked DNA strategy, the phage DNA vaccine can induce more effective immune protection.
