Phage Enrichment from Soil Environment

Phage Enrichment from Soil EnvironmentsBacteriophages are very abundant in soil, and the direct counting method found that there are 107-109 phages per gram of soil. Viruses are also very abundant in fresh water and seawater, which may be close to 107 per milliliter. The global number is estimated to be 1031. However, certain phages are sometimes difficult to isolate directly, so enrichment procedures are usually required. Unfortunately, this technique will limit the diversity of isolated phages, because any enrichment scheme will lead to bias, that is, phages that can reproduce most efficiently under experimental conditions dominate. At Creative Biolabs, we can build different bacterial models to enrich the phages from the soil. For example, we used Bacillus as a model bacteria to enrich phages from the soil. Our team has advanced technology and rich experience and can provide customers with the most suitable solutions to meet your project needs.

Our Services

The phages enrichment from soil services we provided can use filtered extracts that do not contain contaminated bacteria as the basis for the growth of indicator bacteria to provide the success rate of isolation. By first removing the soil bacteria, the target phage is not hindered by competition with the natural soil bacteria present in the original soil sample. This enrichment method has produced dozens of unique phages from several different soil types, and even different types of phages from the same enriched soil sample isolate.

The Workflow of Phage Enrichment from Soil

  • Preparation of bacteria models for phage Isolation
    • Culture Arthrobactersp
    • Allow approximately 24 hours of growth to obtain late index/early stable cells for phage infection experiments
  • Collection of phage from soil samples
  • Collect 200-400 grams of soil from the desired location.

  • Phage Isolation
  • Our phage isolation methods include direct isolation of phage and enriched isolation of phage.

  • Phage Purification
  • Our phage purification methods include PEG precipitation, chloroform extraction, anion-exchange chromatography and size-exclusion chromatography.

  • Results analysis
    • Plaque titer assay
    • Electron micrographic are used for the enrichment technique
    • The stripe plate technique are used to separate a single plaque
    • Isolation of plaques of one phage at different temperatures
