Phagemids and Helper Phages Dual-genome Display System Construction

There are two types of vectors commonly used for phage display: phage vectors and phagemid vectors. Phagemid vectors are currently the most commonly used because of the advantages of smaller genomes, easier manipulation, larger insertable fragments, higher transformation efficiency, and more stable recombinants. However, phagemid vectors only carry the coat protein gene fused to the cDNA of the protein of interest. Therefore, after the transformation of a bacterial host with a phagemid, it requires co-infection from a helper phage to provide native genes for all other structural proteins to form a fully functional phage. The genome of the helper phage lacks packaging signals, so the genome of the phagemid vector will be preferentially packaged into new phage particles, preserving the link between the phenotype and genotype of the phage. Both libraries building and libraries screening require helper phages to provide the proteases and coat proteins required for replication and packaging of the host cell's phagemid DNA.

  • Helper phage containing full-length gIII gene
  • The helper phages containing full-length gIII mainly include M13KO7, R408 and VCSM13, which are the most widely used helper phages and are often referred to as standard helper phages.

  • Helper phage with partial deletion of the gIII gene
  • Deletion of the open reading frame of the gIII gene from the helper phage M13KO7 genome.

  • Helper phage with complete deletion of the gIII gene
  • Helper phages M13MDD3.2, R408d3, and VCSM13d3 are constructed by deleting the gIII gene of helper phages M13KO7, R408, and VCSM13, respectively.
