Many studies have confirmed that ultraviolet radiation can produce a variety of active oxygen. They are very active, have strong oxidizing properties, and can cause damage to biological macromolecules such as DNA and proteins. For example, ultraviolet light can cause many forms of DNA damage. Irradiating the lysogen with an appropriate dose of ultraviolet light can effectively induce the prophage to enter the lysis pathway. At Creative Biolabs, this study established two new methods, namely the colony counting method and the plate induction method, to determine the ultraviolet induction rate of the prophage. The lysogen cultured in the dark after induction by ultraviolet irradiation was directly spread on the plate, and the induction rate of prophage is calculated by CFU (colony forming unit). A plate containing a mixture of lysogen and indicator bacteria is induced by ultraviolet irradiation and calculation of the induction rate of prophage is based on PFU (plaque-forming unit). These two methods can not only accurately determine the frequency of phage induction, but are also simple to operate, labor-saving and instrument-saving, and easy to reuse.
