T4 Phage Display System Construction

T4 phage display system is a new display system established in the mid-1990s. It is worth noting that it can fuse two exogenous peptides or proteins with completely different properties with the coat proteins smaller outer capsid (SOC, 9 ku) and highly immunogenic outer capsid (HOC, 40 ku) on the surface of the T4 capsid, and directly on the surface of the T4 phage Display. Therefore, the expressed protein does not require complicated protein purification, which avoids protein denaturation and loss caused by purification. Because HOC and SOC are non-essential accessory proteins in the structure and life history of phage, their presence or absence does not affect the survival and reproduction of T4, so HOC and SOC have become the key group that can be genetically manipulated in the T4 phage display technology system. One or two exogenous polypeptides (proteins) can be fused to the C-terminus of SOC and/or fused to the N-terminus of HOC and displayed on the surface of T4 phage, but it does not affect the normal activity of T4 phage.
