Phage-Derived Enzymes Production

Some phage-encoded enzymes may help treat bacterial infections and biofilms. These phage-derived enzymes or enzyme biogenesis can be used as an alternative to antibiotics to promote human and animal health. Their efficacy has been demonstrated in some preclinical studies, but these products are still in development. Under current safety standards and regulations, the application of bacteriophage products is easier than using the bacteriophages themselves. Two main types of phage-degrading enzymes can be used to remove biofilms: lysins and depolymerases. Lysin has bactericidal effect on sensitive bacteria. They break peptidoglycan bonds and degrade bacterial cell walls and biofilm structures. Depolymerases can facilitate the early stages of phage infection by degrading extracellular material that encapsulates bacteria and may also help reach phage receptors. Depolymerase activity is particularly interesting in removing biofilms as it alters the EPS matrix and reduces bacterial virulence. Phage depolymerases serve as an alternative to antibiotics for the treatment of multidrug-resistant bacteria, and not only can they be engineered to increase their degradative activity, but they can also be used to detect bacteria.

Our Technology

In order to extract proliferating phages from host microorganisms during production, cell lysis must be performed, which results in the growth medium being saturated with nucleic acids (DNA and RNA) and intracellular components, making the solution viscous and very difficult to handle and purify.
