Receptor and Ligand IdentificationMembrane receptors are essential for the intercellular biochemical and electrical signal transduction of basic physiological functions. Therefore, the pharmaceutical industry tends to focus on the development of drugs that target membrane receptors. Phage display technology provides a rapid method by which proteins and peptides that specifically bind to predetermined molecular targets can be isolated from extremely complex combinatorial libraries. Phage display can advance receptor-based research in many important ways. In the phage display strategy, as many as 109 differently arranged peptides are expressed on the surface of the filamentous phage. Therefore, peptides that can bind to target molecules in vitro and even target tissues in vivo can be identified. In recent years, a series of libraries displaying degenerate peptides of different lengths have been constructed, and specific ligands for cell surface receptors, such as integrins, have been isolated. After biopanning in vivo and intravenous injection of phage libraries into mice, peptides targeting different organs or tumors have been rescued. In one application, isolated peptide ligands have been used to direct cytotoxic drugs to the tumor vasculature of mice.
