Phage Isolation and Purification

Before studying any bacteriophage or using its biological properties, it must first be separated and purified. Therefore, in many explorations of phage biology and biotechnology, separation and purification are the key steps. For a phage to be used for phage therapy, it must be isolated from the environment and exhibit certain characteristics, not just a strain that kills the target bacterial pathogen. These include desirable characteristics, such as a relatively broad host range and lack of other characteristics, as well as the ability to carry toxin genes and form lysogens. Although bacteriophages are usually isolated first and then characterized, the isolation procedure can be changed to bias the isolation towards phages with the desired characteristics. There are several classical and modern techniques to isolate bacteriophages, and the selection of the appropriate method usually depends on the intended use of the bacteriophages. The Methods of Phage Isolation and Purification

Method Description Advantages
Spot testing The plate is inoculated with host bacteria to form a lawn, and then small drops of phage filtrate are placed on the surface. The lysis zone after incubation indicates the presence of phage. It is simple and allows multiple phage filtrates to be tested on the same plate.
Plaque testing The increasingly diluted phage filtrate is mixed with the bacteria and placed on the surface of the plate by spreading or covering with soft agar. It can prove effective phage growth.
Culture lysis The phage filtrate is added to the broth culture of the bacteria and incubated. it is useful for bacteria that will not grow to confluence on solid media.
Routine test dilution The phage lysate is diluted to the extent that it produces less than confluent lysis on the plate. It is suitable for phages that do not form obvious plaques or very small fuzzy plaques.
