Phage Display Library Construction

Phage display technology is an in vitro screening technology used to identify ligands for proteins and other macromolecules. The key to phage display technology is the ability to express peptide or protein sequences as fusions with phage coat proteins. The phage-displayed peptide or protein library is thus physically linked to its encoding nucleic acid, allowing the selection of numerous target types of binding partners through iterative rounds of in vitro panning and amplification, followed by DNA sequencing. A library of more than one billion members can be screened within a few days, providing an effective alternative to more traditional epitope mapping, receptor-ligand identification, or protein evolution methods. Phage display technology relies on the generation of millions of phage libraries. Phage display library technology refers to cloning and expressing the variable region genes of all antibodies of a certain animal in a plasmid or phage, and then screening clones carrying specific antibody genes through different antigens to obtain corresponding specific antibodies or polypeptides.
